Yes it would seem that this cannot be edited via the UI, I can confirm it can be edited via Force.com IDE. Or you can just recreate it manually, which maybe easier if your not using Force.com IDE already.

I managed to solve the situation. I went to my custom object's tab, on the " Buttons, Links and Actions". I went to the tab line and clicked "Edit". And from there, I was able to override the standard page. Note : The page used when overriding a tab shouldn't use a standard controller.

I encountered same issue. And the solution which worked for me was that while creating the permission set I was not selecting any User License in the picklist. I created a new permission set with User License as 'Salesforce' and then option for Tab settings in opportunity object became enabled. May be this could help.
