1) I go to Edit Page, Page Layout editor -> Releated Lists and drag the 'Work Orders' related page to the layout section.

2) Click on Preview as and the Related Work Orders is showing up fine.

3) Login as the same preview User, but now the Related List is not shown on the lightning layout. We are not using Classic Version.

BTW, the Object can be viewed in a separate tab, but just not showing up in the related list.

This is strange behaviour and I'm mainly guessing but maybe I'll be correct.

1st guess:

You have multiple record types/page layouts and the one you're editing is not the one that is assigned to the record type of the record you're viewing later.

Go to page layout assignments in page layouts of object and check this.

2nd guess (but it's not so probable as the first one):

Salesforce likes to catch a lot of things. Maybe combo of browser + salesforce cached old view. Use shift+f5 in a browser and check if page changes.
